TCID50 assays are being used to determine the infectious titer of any such virus types, which can cause cytopathic effects (CPE, structural changes in a host cell resulting from viral infection) in tissue culture over a reasonable period of 5 to 20 days while cells in culture remain viable.
TCID50 (50% tissue culture infectious doses) signifies the concentration at which 50% of the cells are infected when a test tube or well plate upon which cultured cells have been inoculated with a diluted solution of viral fluid. The TCID50 assay is one of the methods for determining the titration of a virus. The TCID50 assay is used to quantify viral titers by determining the concentration at which 50% of the infected cells display cytopathic effect. This method relies on the morphological change of the cell when it has been infected by the virus. The procedure is performed to determine the infectious titer of any virus that can cause cytopathic effects in tissue culture and is carried out for 5 to 20 days in culture to remain viable. CPE occurs when the infecting virus causes lysis (dissolution) of the host cell or when the cell dies without lysis because of its inability to reproduce. The degree of CPE can indirectly reflect the virulence of the virus, so this feature can be used to determine the virulence of the virus. It should be noted that not all viruses can cause CPE in tissue culture. Cell lines and viruses should be aligned to see a cytopathic effect.
