Flaviviruses Antiviral Services

The genus Flavivirus belongs to the Flaviviridae family and consists of more than 70 members, including the prototype yellow fever virus (YFV), four serotypes of dengue virus (DENV1, DENV2, DENV3, and DENV4), West Nile virus (WNV), Kunjin virus (KUN), tick-borne encephalitis virus (TBEV), and Japanese encephalitis virus (JEV). Flaviviruses are single-stranded positive-sense RNA viruses whose genomes encode 3 structural proteins (capsid (C), membrane (M), and envelope (E)) and 7 nonstructural proteins. Among them, NS3 protease and its cofactors NS2B and NS5 are the most studied antiviral interventions. The NS5 protein contains the methyltransferase/guanylyltransferase (MTase/GTase) domain for mRNA capping and the RNA-dependent RNA polymerase (RdRp).

Flavivirus infection is a global threat and is endemic or epidemic in almost every tropical country. Even with reduced morbidity and mortality due to the use of various vaccines, outbreaks of Flaviviruses are still occurring, highlighting the challenges in implementing effective methods to control outbreaks. To date, there are no specific commercial drugs against Flaviviruses, so there is an urgent need for new drugs that can be used for prophylactic or therapeutic interventions against Flavivirus infections.

Our in vitro testing services include viral screening tests as well as tests to evaluate promising candidates, including their range of action, the effectiveness of drug combinations, the potential for resistance selection, and phenotypic/genetic characterization of resistant strains. For in vivo efficacy studies, we provide in vivo model development and testing services. High-potential candidates can be rapidly identified through our screening assay, with simultaneous efficacy and cytotoxicity assessments. Depending on the desired viral model, we can use a variety of cell lines including Vero, BHK21, LLCMK2, and Huh7. Analysis can be performed using primary cells and can be customized on request. Creative Diagnostics can also perform customized neutralization assays.
